Edit your question to include the IP of the iSCSI target, the output of tracert to that IP, and the output of route print.

How is your iSCSI initiator configure? Could you share the details?
You can use Wireshark or similar tools. Filter via iSCSI protocol to analyze network traffic. https://wiki.wireshark.org/iSCSI
In addition, you can simply run diskspd benchmark and monitor network traffic in the Task Manager.

As an alternative, you can use StarWind VSAN to create iSCSI Target. Using ACL you can bound traffic to a specific NIC.
