Yes you can, but in general you shouldn't. How well this performs will depend on various factors, such as concurrent user count, usage of backend services (i.e. SharePoint 2010 Workflows), etc.
so in that case do we need to select/define WFE & app role separately while cofigurig min role?

You could use custome role to combine Appplication and Web Front-End.
