May I ask what would be the behavior if ever the RODC ( Read only domain controller) on other site will be affected just in case we upgrade to new ADDS 2019?

The two prerequisites to introducing the first 2019 domain controller are that domain functional level needs to be 2008 or higher and older sysvol FRS replication needs to have been migrated to DFSR

No effect.
Upgrading the DC will not affect the running RODC
