Yes, you could Upgrade Windows Server 2012 R2 to Windows Server 2019, but it may lead too unexcepted issue, users usually don’t do this way.

In my opinion, I prefer to clean install Windows Server 2019 and start a fresh WSUS role install, then sync with Microsoft Update, create your groups, adjust your settings, and then change GPOs over to the new server.
Clean system and new install will make all things smoothly.
