I haven't found any information if it is possible running 2 MSMQ roles on single server/cluster. However, you can still deploy multiple VMs on top of your cluster with MSMQ roles running inside. The following article should help with MSMQ role configuration within the cluster:

I have not tried it but you can have Two MSMQ roles in one Failover cluster manager. Microsoft Message Queuing (MSMQ) stores emails temporally, while they are scanned or waiting to be actioned by the mail server. So I suppose there is no need for two MSMQ but still, it can be done.
