All our laptop joined AAD and using UPN to sign in, and after first sign in, users will have to set an alternative sign in method which is using pin. If the UPN/sing in is blocked, can the user still able to sign in the laptop using hello pin?

Windows Hello for Business PIN or password doesn't matter for this, blocking account sign in in AAD does not prevent a user from logging into a Windows endpoint that is AAD joined as there is no communication to AAD that happens at the time the local login happens.
