Based on my research, I find that we can use conditional access policies to require multi-factor authentication (MFA) for device enrollment to help you secure your corporate resources.

With add "Microsoft Intune Enrollment" in the conditional access policy's Cloud apps and select "Require multi-factor authentication" in Grant access, MFA will pop up during enrollment and login of the company portal app. For more details, please refer to the following article.

In conclusion, based on my understanding, ios supports MFA in DEP Enrollment(now named "ADE enrollment").
