I have an ISO file provided by internal licensing team, they have confirmed that it is for Core based licensing. Now can I use the same ISO file for one of the server where we want it to be CAL based.
Please confirm.

The license is driven by the product key, not by the installation media.
You could even download the Evaluation Edition to install it with your product key.
It depends on the version of SQL Server. Older versions had different media based on licensing. Since SQL 2016, they are all the same media, controlled by the product key.
