I checked and found that our server doesn't have "Temporary ASP.NET Files" in the folder v3.5. However our application pool are connected to v2, so I checked path "C:\Windows\Microsoft.NET\Framework\v2.0.50727\Temporary ASP.NET Files" and yes, that folder exists. Also I found that IIS_IUSRS already has access of everything except for "Full control". (see attached screen shot)
Also regarding Step 2, we do have few of the applications running on MVC and requires .net framework 4 also, so wanted to know should I still go ahead delete all files under folder "C:\Windows\Microsoft.NET\Framework\v4.0.30319\Temporary ASP.NET Files"? Please let us know.

Hi @SamWu-MSFT ,
Reopening this issue, we were able to resolve this issue if we delete temporary files. But again after 10-15 days, it has started created issue again.
Also I have gone through the link you provided, we are having this error with application having .Net Framework 3.5, so that link will not be useful for us.
Help us with any other solution.

Hi @SamWu-MSFT , I added one user and provided custom account for that pool identity, however it is still throwing error.
Also I had done aspnet/regiis.exe -ga "ThatCUSTOMUSER".
But still it is throwing same error.

Hi @SamWu-MSFT, I am not able to find that DLL in the folder you mentioned. Regarding Identity, we are not using Network Service, but are using the "ApplicationPoolIdentity"
You may find the same in attached screenshot. Let me know what can I do to resolve this issue? Appreciate your help in advance.
